Морфологические изменения и экспрессия каспаз в почках крупного рогатого скота в возрастном аспекте
Аннотация
Обоснование. Ввиду высокой метаболической нагрузки почки коров особенно подвержены воздействию факторов промышленного содержания и претерпевают морфологические изменения, проявляющиеся в том числе гибелью клеток. В современной литературе не представлено данных по изучению ферментов, играющих важную роль в апоптозе клеток почек крупного рогатого скота разных возрастных групп. В настоящей статье описаны результаты морфологического анализа и иммуногистохимического окрашивания на выявление каспазы-3, -6, -8 в почках крупного рогатого скота разных возрастных групп.
Цель – оценить возрастные морфологические изменения в почках крупного рогатого скота и их взаимосвязь с экспрессией каспаз.
Материалы и методы. Работу выполняли в Уральском НИВИ – структурном подразделении ФГБНУ УрФАНИЦ УрО РАН. Для изучения морфологических изменений почек постмортально был отобран материал от крупного рогатого скота следующих возрастов: телята 2,5–10 месяцев; коровы 2–5 лет; коровы 6–8 лет. Проводили иммуногистохимическое окрашивание образцов с антителами к каспазам-3, -6, -8 для оценки интенсивности апоптоза в органе. В почках измеряли количество и площадь почечных телец, площадь почечного клубочка и пространства капсулы Боумена-Шумлянского, долю площади, занимаемую ими в почечном тельце, а также долю поврежденных почечных телец.
Результаты. Развитие патологических процессов в почках крупного рогатого скота начинается в раннем возрасте. Морфометрический анализ препаратов почек выявил прогрессирующие с возрастом патологические нарушения, ассоциированные с высоким уровнем экспрессии каспаз, особенно исполнительных – каспазы-3 и -6.
