Пробиотические свойства молочнокислых бактерий, выделенных из репродуктивной системы коров экоферм Северо-Запада России
Аннотация
Обоснование. Применение пробиотиков, способных восстанавливать нормальную микробиоту репродуктивной системы коров и повышать устойчивость к инфекциям является многообещающим направлением.
Целью исследования было выделение, идентификация и характеристика перспективных штаммов молочнокислых бактерий, выделенных из репродуктивного тракта здоровых коров, для разработки нового биопрепарата, направленного на профилактику эндометрита.
Материалы и методы. Биологический материал (соскобы со стенок влагалища) для выделения перспективных штаммов лактобактерий отбирался у 10 клинически здоровых дойных коров из трех экоферм (Северо-Западного региона РФ и республики Карелии). Выделение чистых культур осуществляли методом посева серийных разведений на селективную агаризованную среду, а идентификацию проводили стандартными микробиологическими методами, включая ПЦР-анализ. Антагонистическую активность против 10 патогенных и условно-патогенных микроорганизмов оценивали in vitro методом диффузии в агар.
Результаты. Выделено 95 изолятов лактобактерий. Скрининг 42 штаммов лактобактерий выявил 19 активных и умеренно активных вариантов с антимикробной активностью в отношении патогенов. Штамм Э-49 (Lactobacillus sp.) показал максимальную антагонистическую активность против Clostridium perfringens (38±1,0 мм) и Enterococcus faecalis (32±1,9 мм). Штамм Э-52 (Lactobacillus sp.) активно подавлял рост Penicillium sp. (37±1,5 мм) и Streptococcus agalactiae (21±0,9 мм). Все протестированные штаммы не проявляли гемолитической активности. Некоторые активные штаммы, такие как Э-49, Э-52 и Э-10, показали стабильные титры жизнеспособных клеток в течение 30 суток хранения (2,4×10⁹ ± 1,5×10⁸ и 1,4×10⁹ ± 7,0×10⁷ КОЕ/мл для Э-52 через 10 и 30 суток, 7,3×10⁹ ± 6,2×10⁸ КОЕ/мл для Э-10 через 30 суток) и оптимальные значения pH (3,8-4,2).
